Effects of Nitrogen on Dry Matter Accumulation and Productivity of Three Cropping Systems and Residual Effects on Wheat in Deep Vertisols of Central India

A field experiment was conducted on deep vertisols of Bhopal, India to study the effects of three levels of nitrogen (N), namely 0, 75 and 100 % of the recommended dose of nitrogen (RDN), on the dry matter accumulation (DMA) and productivity of three cropping systems (sole soybean, sole sorghum and soybean + sorghum intercropping) during the rainy season and their residual effect on the subsequent wheat crop during the post-rainy season. During the rainy season, sole sorghum was found to have significantly higher DMA and productivity in terms of soybean equivalent yield (SEY) than sole soybean or soybean + sorghum intercropping. Increasing the N dose from 0 to 100 % RDN significantly improved the DMA and SEY. At a low fertility level (N0), soybean + sorghum intercropping was found to be more productive, while at a high fertility level (100 % RDN), sole sorghum was more productive than the other two cropping systems. However, during the post-rainy season, sole soybean as the preceding crop gave the highest DMA and seed yield of wheat, which were similar to those found with soybean + sorghum intercropping. Sorghum followed by wheat gave the lowest DMA and seed yield of wheat. Application of 100 % RDN irrespective of cropping system during the preceding crop improved the DMA of wheat but not its seed yield. However, N applied to the wheat crop significantly increased its DMA and seed yield. Einfluss von Stickstoff auf Trockenmasseakkumulation und Produktivität von drei Anbausystemen und deren Rückstandswirkung auf Weizen in einem tiefen Vertisol Zentralindiens Ein Feldexperiment wurde durchgeführt auf einem tiefen Vertisol bei Bhopal, Indien, um den Einfluss von drei Stickstoffkonzentrationen 0, 75 und 100 % der empfohlenen Stickstoffmenge (RDN) auf die Trockenmasseakkumulation (DMA) und Produktivität von drei Anbausystemen (Reinanbau Sojabohne, Reinanbau Sorghum und Sojabohne + Sorghum Mischanbau) während der Regensaison und deren Nachwirkungen auf den folgenden Anbau von Weizen während der Nachregensaison zu untersuchen. Während der Regensaison war der Reinanbau von Sorghum signifikant höher in DMA und in der Produktivität in Form von Ertragsäquivalenten für Sojabohnen (SEY) im Vergleich zu einem Reinanbau von Sojabohne oder einem Mischanbau von Sojabohne + Sorghum. Eine Erhöhung der N-Anwendung von 0 bis 100 % RDN erhöhte Signifikanz DMA und SEY. Unter der niedrigen Düngung (N0) erwies sich Sojabohne + Sorghum Mischanbau als produktiver im Vergleich zu einer hohen Düngungeranwendung (100 % RDN), Reinanbau war produktiver als die anderen beiden Anbausysteme. Allerdings während der Nachregensaison erwies sich der Reinanbau von Sojabohnen vor Weizen als die höchste DMA und Ertragsmenge, was mit dem Sojabohnen + Sorghum Mischanbau vergleichbar war. Sorghum gefolgt von Weizen ergab den geringsten DMA und niedrigsten Weizenertrag. Die Anwendung von 100 % RDN erhöhte unabhängig von dem Anbausystem der vorhergehenden Kulturpflanzen DMA von Weizen aber nicht den Kornertrag. Accounting for residual effects of previously applied nitrogen fertilizer on intensively managed grasslands

Abstract Only 0·20,0·70 of the fertilizer-nitrogen (N) applied to grassland is taken up in herbage in the harvest directly following application. Residual effects at subsequent harvests can be large but are poorly quantified, and rarely taken into account in current management practices. An increased understanding of N-use efficiency per harvest can improve operational management. This study systematically assessed the residual effects of previously applied N fertilizer on N uptake, dry matter (DM) yield and soil mineral-N (SMN) during the whole of the growing season. It is based on field experiments conducted on peat and mineral soils in 1991,1994. Statistical models were derived for SMN, N uptake and DM yield as a function of previously and freshly applied N fertilizer. There were clear residual effects of previously applied N in later cuts. They were relatively greater at higher levels of N fertilizer. On peat soils, 0·15,0·25 of the N applied was recovered as SMN. On mineral soils the proportion was maximally 0·08. There was a clear relationship between SMN and N uptake in the subsequent cut on mineral soils but not on peat soils. The value of SMN as a tool to adjust fertilizer-N application rates was hence found to be limited. There were clear relationships between the amount of previously applied N and the N uptake in subsequent cuts, on both soil types and over the whole of the growing season. It was concluded that the total amount of previously applied N is a useful indicator for adjusting N-fertilizer application rates. [source]


Effects of plant invasions on the species richness of abandoned agricultural land

While exotic plant invasions are thought to lead to declines in native species, the long-term impacts of such invasions on community structure are poorly known. Furthermore, it is unknown how exotic plant invasions compare to invasions by native species. We present data from 40 yr of continuous vegetation sampling of 10 fields released from agriculture to examine the effects of invasions on species richness. The effects of both exotic and native species invasions on species richness were largely driven by variations among fields with most species not significantly affecting species richness. However, invasion and dominance by the exotics Agropyron repens, Lonicera japonica. Rosa multiflora. Trifolium pratense and the native Solidago canadensis were associated with declines in richness. Invasions by exotic and native species during old field succession have similar effects on species richness with dominance by species of either group being associated with loss of species richness. Exotic species invasions tended to have stronger effects on richness than native invasions. No evidence was found of residual effects of invasions because the impact of the invasion disappeared with the decline of the invading population. When pooled across species, heavy invasion by exotic species resulted in greater loss o species richness than invasion by native species. Studies of invasion that utilize multiple sites must account for variability among sites. In our study, had we no included field as a factor we would have incorrectly concluded that invasion consistently resulted in changes in species richness. [source]


Craving: what can be done to bring the insights of neuroscience, behavioral science and clinical science into synchrony

Alcohol self-administration behavior is the common thread that is necessary to bring the insights of neuroscience, behavioral science and clinical science into synchrony around the concept of craving. Animal models should address the molecular and cellular changes that take place in behaviorally relevant brain regions of rats consequent to chronic self-administration of ethanol. Animal models can focus on the biology of the anticipatory state in alcohol preferring/consuming rats, as well as studies of the effects of possible medications on this state in the animal model, on actual alcohol consuming behavior, and on the residual effects of chronic alcohol on the non-human mammalian brain. In human studies of craving, cue-reactivity in the absence of the opportunity to drink alcohol does not have the same salience as cue-reactivity in which drinking is possible. Moreover, actual drinking behavior serves to validate self-reports of craving. Studies of limited alcohol self-administration in the laboratory are an essential element in screening new medications for the treatment of alcoholism. Studies to date suggest no adverse reaction to the participation of alcoholic subjects in limited alcohol self-administration studies, but the research community should continue to monitor carefully the outcomes of alcohol-dependent subjects who participate in this type of research, and efforts should always be made to encourage these subjects to enter active treatment. In outpatient clinical trials of new treatments for alcoholism, the assessment of craving should include queries regarding symptoms and signs of protracted abstinence such as sleep disturbances, as well as questions regarding situational craving. Field observations of alcoholics in their favorite drinking environments would contribute greatly to our understanding of the real-world phenomenology of craving. [source]


Loss of zolpidem efficacy in the hippocampus of mice with the GABAA receptor ,2 F77I point mutation

Abstract Zolpidem is a hypnotic benzodiazepine site agonist with some ,-aminobutyric acid (GABA)A receptor subtype selectivity. Here, we have tested the effects of zolpidem on the hippocampus of ,2 subunit (,2F77I) point mutant mice. Analysis of forebrain GABAA receptor expression with immunocytochemistry, quantitative [3H]muscimol and [35S] t-butylbicyclophosphorothionate (TBPS) autoradiography, membrane binding with [3H]flunitrazepam and [3H]muscimol, and comparison of miniature inhibitory postsynaptic current (mIPSC) parameters did not reveal any differences between homozygous ,2I77/I77 and ,2F77/F77 mice. However, quantitative immunoblot analysis of ,2I77/I77 hippocampi showed some increased levels of ,2, ,1, ,4 and , subunits, suggesting that differences between strains may exist in unassembled subunit levels, but not in assembled receptors. Zolpidem (1 µm) enhanced the decay of mIPSCs in CA1 pyramidal cells of control (C57BL/6J, ,2F77/F77) mice by ,,60%, and peak amplitude by ,,20% at 33,34 °C in vitro. The actions of zolpidem (100 nm or 1 µm) were substantially reduced in ,2I77/I77 mice, although residual effects included a 9% increase in decay and 5% decrease in peak amplitude. Similar results were observed in CA1 stratum oriens/alveus interneurons. At network level, the effect of zolpidem (10 µm) on carbachol-induced oscillations in the CA3 area of ,2I77/I77 mice was significantly different compared with controls. Thus, the ,2F77I point mutation virtually abolished the actions of zolpidem on GABAA receptors in the hippocampus. However, some residual effects of zolpidem may involve receptors that do not contain the ,2 subunit. [source]


Absence of residual effects of a defeated resistance gene in poplar

Summary In a few plant pathosystems, defeated major genes have been shown to contribute to partial resistance to disease. This hypothesis has never been tested before in a forest tree, but pathogenic variation associated with recent hybridization in poplar rust in the Pacific northwest provided an opportunity. An F2 progeny of 256 poplar clones in the field near Corvallis, Oregon, USA, has been monitored for rust severity and infection type since the advent of the new hybrid rust, Melampsora × columbiana, in the mid-1990s. All 256 clones displayed a susceptible infection type in 1997 and again in 2000, and yet variation in uredinial density (i.e. partial resistance) was still observed. To determine which clones possessed a defeated resistance gene, a greenhouse inoculation was performed with an isolate of M. medusae, one of the parents of M. × columbiana. Clones that would have been resistant to M. medusae, prior to the advent of M. × columbiana, were thus identified. The inoculation resulted in a 1 : 1 segregation (,2=0.772; p=0.38) for resistance, indicating the presence of a major gene. However, the F2 clones possessing the defeated resistance gene displayed the same level of partial resistance in the field in both 1997 and 2000 as their full siblings lacking the gene. Résumé Chez quelques pathosystèmes végétaux, il a été montré que le contournement de gènes majeurs de résistance contribue à une résistance partielle envers la maladie. Cette hypothèse n'a encore jamais été testée chez un arbre forestier, mais le changement de pouvoir pathogène associéà l'hybridation récente de la rouille du peuplier dans le nord-ouest des USA en a fourni l'occasion. Une descendance F2 de 256 clones de peuplier a été suivie au champ près de Corvallis, Oregon, USA, pour la gravité de la rouille et le type d'infection, depuis l'apparition du nouvel hybride Melampsora x columbiana, dans les années 1990. Tous les 256 clones se sont montrés sensibles en 1997 et à nouveau en 2000, et une variation dans la densité des urédies (résistance partielle) a aussi été observée. Für Waldbäume wurde diese Hypothese bisher nie überprüft. Dies wurde jetzt im pazifischen Nordwesten möglich, wo der Pappelrost nach einem rezenten Hybridisierungsereignis stark variierte. An den F2-Nachkommenschaften von 256 Pappelklonen, die unter Freilandbedingungen in der Nähe von Corvallis, Oregon, USA wuchsen, wurde nach dem Auftreten des neuen Hybridrostes (Melampsora × columbiana) ab ca. 1990 die Krankheitsintensität und der Infektionstyp registriert. Alle 256 Klone zeigten einen anfälligen Infektionstyp im Jahre 1997 und dann wieder im Jahre 2000. Dabei wurde eine Variation in der Urediendichte (d.h. partielle Resistenz) beobachtet. Effects of 15N Split-application on Soil and Fertiliser N Uptake of Barley, Oilseed Rape and Wheat in Different Cropping Systems

Abstract In intensive farming systems, farmers split up and apply the N fertilization to winter cereals and oilseed rape (OSR) at several dates to meet the need of the crop more precisely. Our objective was to determine how prior fertilizer N application as slurry and/or mineral N affects contributions of fertilizer- and soil-derived N to N uptake of barley (1997), oilseed rape (OSR; 1998) and wheat (1999). In addition, residual fertilizer N effects were observed in the subsequent crop. Since autumn 1991, slurry (none, slurry in autumn, in spring, in autumn plus in spring) and mineral N fertilizer (0, 12 and 24 g N m,2) were applied annually. Each year, the treatments were located on the same plots. In 1997,1999, the splitting rates of the mineral N fertilization were labelled with 15N. Non-fertilizer N uptake was estimated from the total N uptake and the fertilizer 15N uptake. All three crops utilized the splitting rates differently depending on the time of application. Uptake of N derived from the first N rate applied at the beginning of spring growth was poorer than that from the second splitting rate applied at stem elongation (cereals) or third splitting rate applied at ear emergence or bud formation (all three crops). In contrast, N applied later in the growing season was taken up more quickly, resulting in higher fertilizer N-use efficiency. Mineral N fertilization of 24 g N m,2 increased significantly non-fertilizer N uptake of barley and OSR at most of the sampling dates during the growing season. In cereals, slurry changed the contribution of non-fertilizer N to the grain N content only if applied in spring, while OSR utilized more autumn slurry N. In OSR and wheat, only small residual effects occurred. The results indicate that 7 years of varying N fertilization did not change the contribution of soil N to crop N uptake. [source]


Assessment of bismuth thiols and conventional disinfectants on drinking water biofilms

Abstract Aims: Biofilms in water distribution systems represent a far more significant reservoir of micro-organisms than the water phase. Biofilms are (i) resistant to disinfectants, (ii) nuclei for microbial regrowth, (iii) a refuge for pathogens, (iv) accompanied by taste and odour problems, and (v) corrode surfaces. The effects of the current strategies for disinfection of drinking water systems in large buildings (chlorination, copper and silver ionization, and hyper-heating) were compared with a new generation of bismuth thiol (BT) biocides. Methods and Results: Multispecies biofilms were treated with 0·8 mg l,1 of free chlorine, 400 and 40 ,g l,1 of copper and silver ions, respectively, at 55 and 70°C, and bismuth-2,3-dimercaptopropanol (BisBAL). Furthermore, the effect of combined heat and BisBAL on planktonic cell viability was examined in monoculture using Escherichia coli suspensions. Inactivation rates for BisBAL were similar to copper,silver ions, where the effects were slower than for free chlorine or temperature. The BisBAL effect on E. coli monocultures was augmented greatly by increasing temperatures. Conclusions: Like copper,silver ions, BTs show more persistent residual effects than chlorine and hyper-heating in water systems. BT efficiency increased with temperature. Like copper,silver ions, BT action is relatively slow. Significance and Impact of the Study: BT presents a new approach to containing water biofilms. BT action is not as rapid, but is more thorough than chlorine, and less caustic. BTs may also be more efficacious in hot water systems. At sub-minimum inhibition concentration levels, BTs uniquely inhibit bacterial exopolysaccharide, thereby retarding biofilm formation. Thus, the combination of bactericidal and residual effects may prevent slime build-up in hot water systems. [source]


Effect of toothpaste on the plaque inhibitory properties of a cetylpyridinium chloride mouth rinse

Abstract Background and aims: Cetylpyridinium chloride (CPC) mouth rinses have moderate plaque inhibitory activity when used alone but rarely have shown adjunctive benefits to tooth brushing with toothpaste. Several explanations for this apparent anomaly can be proferred, including loss of antiseptic activity due to interactions with toothpaste ingredients. The aim of this study was to measure the effect of toothpaste on the plaque inhibitory properties of a CPC mouth rinse using paired rinses of CPC, toothpaste slurry (TP) and water (W). Methods: The study was a single blind, randomised, seven-treatment, cross over design balanced for residual effects, involving 21 healthy, dentate subjects. The paired rinses were: W-CPC, CPC-W, TP-CPC, CPC-TP, W-TP, TP-W and W-W. Rinsing with solutions or slurries was done for 60 s twice per day. On day 1, subjects were rendered plaque free, suspended tooth cleaning and commenced the allocated rinse regimen. On day 5, plaque was scored by index. A 2½ day wash out of normal oral hygiene was allowed between each regimen. Results: The order from lowest to highest plaque scores was as follows: W-CPC = CPC-W < CPC-TP < TP-CPC , W-TP < TP-W < W-W. Several differences in pairs of treatments were statistically significant, the most relevant of which were significantly less plaque with W-CPC compared to TP-CPC, TP-W and W-TP, and significantly more plaque with W-W compared to all other regimens except TP-W. Conclusions: Toothpaste, whilst possessing some plaque inhibitory activity, when used immediately before a CPC mouth rinse adversely affected the plaque inhibitory action of this antiseptic. This in part may explain the reported lack of adjunctive benefits of CPC rinses to normal oral hygiene practices and supports the suggestion, made for chlorhexidine rinses, that their use should follow toothpaste by at least 60 min. Es wurde jedoch selten gezeigt, dass sie zusätzlich zum Zähneputzen Vorteile haben. Mehrere Erklärungen für diese offensichtliche Anomalie können angeboten werden, einschließlich des Verlusts der antiseptischen Aktivität wegen einer Interaktion mit Inhaltstoffen der Zahnpasta. Ziel dieser Studie war es, die Wirkung der Zahnpasta auf die plaquehemmenden Eigenschaften einer CPC-Mundspüllösung zumessen. Zwischen den Spülmaßnahmen war eine 2,5-tägige Auswaschperiode mit normaler Mundhygiene erlaubt. Above- versus below-ground competitive effects and responses of a guild of temperate tree species

Summary 1The neutral theory debate has highlighted the scarcity of robust empirical estimates of the magnitude of competitive effects and responses within guilds of co-occurring tree species. Our analysis quantifies the relative magnitude of all possible pairwise competitive interactions within a guild of nine co-occurring tree species in temperate forests of northern, interior British Columbia, and explicitly partitions the competitive effects of neighbours into the effects of shading versus the residual effects of ,crowding', assumed to reflect below-ground competition. 2Models that treated neighbours as equivalent in their competitive effects were the most parsimonious for the five species with the smallest sample sizes. For the remaining species (samples sizes of > 150 individuals), the best models estimated separate competition coefficients for all nine species of neighbours. We take this as evidence that species do indeed differ in their competitive effects, but that there can be a minimum sample size required to discriminate between them. 3There was a strong size-dependency in potential growth. Six species showed an optimal growth at a size between 5 and 20 cm diameter. Potential growth declined moderately to strongly as diameter increased. Sensitivity to crowding varied as a function of tree size for five of the nine species; however, this response was not consistent by tree species. 4The magnitude of reduction in growth due to crowding was greater on average than the reduction in growth due to shading, except for the two least shade tolerant conifers. Sensitivity to shading among the conifer species was correlated with their shade tolerance. 5The per capita effects of crowding by different species of neighbours varied widely. A large number of the estimated pairwise per capita competition coefficients were very low. The relative magnitude of the strength of intra- versus interspecific competition also varied widely among the tree species. 6Synthesis. Model selection techniques effectively separated above- and below-ground competition in complex forests, and allowed us to assess differences among species in competitive effects and responses. While below-ground effects were strong, they were due to proximity of neighbours from a very specific (and small) subset of strong competitors within the guild. Response to crowding varied with tree size but the nature of the relationship varied widely among the species. [source]

Summary Gaboxadol is a selective extrasynaptic GABAA receptor agonist previously in development for the treatment of insomnia. Due to its short half-life (1.5,2 h) it is expected to be free from residual effects the next morning. The present study assessed the residual effects of evening and middle-of-the-night administration of 15 mg of gaboxadol on cognitive, psychomotor and driving performance. Twenty-eight healthy volunteers entered the study with 25 (12 women; mean age 31.4 years) completing a double-blind, placebo-controlled, active-referenced five-way cross-over study. Each treatment night subjects ingested one capsule at 23:00 hours and one at 04:00 hours. Treatments were placebo at both times, 15 mg gaboxadol or 7.5 mg zopiclone followed by placebo, and placebo followed by 15 mg gaboxadol or 10 mg zolpidem. Effects on cognition and psychomotor performance were assessed between 07:30 and 08:30 hours and on driving between 09:00 and 10:00 hours. Driving, as measured by standard deviation of lateral position in an on-the-road driving test, was almost significantly (P < 0.07) impaired after evening administration of gaboxadol for the all-subjects-completed set (n = 25) but significantly (P < 0.05) in the full analysis set (n = 28). Effects of all other active treatments on driving were significant. Evening administration of gaboxadol had minor effects on divided attention only, whereas middle-of-the-night administration impaired performance significantly in all tests except memory. Zolpidem and zopiclone impaired performance significantly in every test except tracking after zopiclone; 15 mg of gaboxadol can produce minor residual effects on driving after evening administration. Administration later at night is associated with moderately impairing residual effects on driving and psychomotor performance but not on memory. [source]


Metabolic response to two hydrocooling temperatures in sweet cherries cv Lapins and cv Sunburst

Abstract Physiological and metabolic characterisation and analysis of response to two hydrocooling temperatures in cv Sunburst (early season) and cv Lapins (mid-season) cherries during post-harvest life has been studied. Samples were hydrocooled with water at 1 °C to reach 6 °C inside the fruit (HC-6C) and 2 °C (HC-2C) inside the fruit. After harvesting, Sunburst samples presented higher respiration rates and lower malic acid and sorbitol contents than Lapins. Glucose and fructose contents were similar in the two varieties. Sunburst control exhibited a higher respiration rate than Lapins and a higher rate of conversion from sorbitol to fructose. The change of glucose and malic acid consumption over 4 days at 20 °C was similar for the two varieties. Hydrocooling reduced respiration and the consumption of respiratory substrates. The residual effect of hydrocooling was especially significant in cherries of both varieties that had been pre-cooled to 2 °C. Hydrocooling delayed loss of skin and pulp firmness, and reduced loss of titratable acid and soluble solid contents over 4 days at 20 °C in both varieties. Hydrocooling to 2 °C checked loss of quality with respect to controls for both varieties after 4 days at 20 °C. Lapins showed better conservation properties than Sunburst under all the experimental storage conditions. Hydrocooling reduced total losses in both varieties, especially in cherries pre-cooled to 2 °C. Hydrocooling also had several residual effects: reduction of the respiration rate and consumption of respiratory substrates, and slowing of loss of quality, particularly for Lapins. Persistence and efficacy of two diatomaceous earth formulations and a mixture of diatomaceous earth with natural pyrethrum against Tribolium confusum Jacquelin du Val (Coleoptera: Tenebrionidae) on wheat and maize

Abstract Laboratory tests were conducted to assess the insecticidal and residual effects of three diatomaceous earth (DE) formulations, Insecto®, PyriSec® and SilicoSec®, against Tribolium confusum Jacquelin du Val on wheat and maize. Quantities of wheat and maize were treated with the above formulations at 500, 1000 and 1500 mg kg,1 and stored at 25 °C and 55% relative humidity (RH). Samples were taken on the day of storage and every 30 days until completion of a 360 day period of storage. Adults of T. confusum were exposed to these samples at 25 °C and 55% RH and the mortality was measured after 24 and 48 h and 7 and 14 days of exposure. Adult mortality was higher on wheat than on maize. At the beginning of the storage period, mortalities after 14 day exposure on maize treated with the highest rate were 60, 63 and 81% for Insecto®, PyriSec® and SilicoSec® respectively, while on wheat the mortality was 100% for all DEs. On the same commodities 360 days after treatment the respective mortality figures for wheat were 99, 98 and 100%, while in the case of maize they did not exceed 7%. Generally, mortality at exposures ,48 h decreased with increasing storage time. Furthermore, although mortality on wheat increased with dose, the increase in DE dose from 1000 to 1500 mg kg,1 resulted in only a small increase in mortality. Thus a DE treatment of 1000 mg kg,1 was shown to provide long-term protection of wheat against T. confusum, although higher DE application rates and exposure intervals are needed for a satisfactory level of protection of maize against this pest. A Multicenter, Placebo-Controlled, Double-Blind, Randomized Study of Efficacy and Safety of Ocinaplon (DOV 273,547) in Generalized Anxiety Disorder

Preclinical studies demonstrated that ocinaplon, a positive allosteric modulator of GABAA receptors, possesses anxiolytic-like actions at doses devoid of the side effects typically associated with benzodiazepines. The aim of this study was to evaluate the effects of ocinaplon in a multicenter, double-blind proof-of-concept trial of male and female outpatients who met DSM-IV criteria for GAD with no coexisting depression, and had a baseline score of ,20 on the Hamilton Scale for Anxiety (HAM-A). Patients with <20% reduction in HAM-A to placebo in a single-blind 7-day run-in period were randomly assigned to treatment with ocinaplon 90 mg t.i.d. (n = 31) or placebo for 28 days (n = 29). Ocinaplon was more effective than placebo in reducing HAM-A scores (P= 0.009). Patients assigned to ocinaplon exhibited a mean improvement of 14.2 points (SE = 2.6) on the total score of the HAM-A scale at the conclusion of the trial, while patients assigned to placebo obtained a mean improvement of 6.3 points (SE = 2.0). A significant (P= 0.023) difference in improvement between ocinaplon and placebo was observed beginning at and continuing from 1-week after the initiation of dosing. The proportion of patients with treatment-emergent adverse events (TEAE) was not statistically significant between ocinaplon and placebo. One serious adverse event (SAE) occurred in the ocinaplon group that was considered possibly related to study medication (icterus following transaminase elevations). The patient had preexisting medical conditions that may have contributed to this SAE. A full recovery was observed with no residual effects. The overall safety profile revealed no patterns of TEAEs, including those effects typically associated with other anxiolytic and/or benzodiazepine compounds, such as sedation. Ocinaplon appears to be a well-tolerated and effective treatment for GAD. It produces a rapid onset of anxiolytic action absent the side effects (e.g., dizziness, sedation) typically reported following anxiolytic doses of benzodiazepines. [source]
